The Markdown Block allows a user to insert text into a SAM using the full capabilities of what is possible with Markdown when published. SAMs use Markdown as it is more secure than HTML. The Settings allow you to work directly in Markdown or by WYSIWYG with Markdown functionality available as a selection. More information and a tutorial on learning Markdown can be found here.
